Efek hipoglikemik fraksi air kulit batang pepolo (bischofia javanica blume) pada mencit jantan galur balb/c dengan metode toleransi glukosa

Diabetes mellitus (DM) is a disease or chronic metabolic disorder with multiple etiologies characterized by high blood glucose levels accompanied by impaired carbohydrate, lipid, and protein metabolism due to insulin function insufficiency. Pepolo stem bark (Bischofia javanica Blume) is one part of the plant that is used as a traditional medicine for DM. Previous studies have proven that the water fraction of pepolo stem bark has better antihyperglycemic activity than the extract, n- hexane, and ethyl acetate fractions. Therefore, this study aims to prove the hypoglycemic effect of the water fraction of pepolo stem bark and determine the effective dose. The glucose tolerance method's research method with a modified carbohydrate load, namely glucose and sucrose (3 g/kg BW). The test animals used were 15 healthy male Balb/C strain mice for each induction method used and divided into five groups, namely the negative control group (Na CMC 0.5%), the positive control group (glibenclamide 0.65 mg /kg BW), and 3 test groups for water fraction of pepolo stem bark with doses of 200, 400 and 600 mg/kg BW. Induction was carried out 30 minutes after the treatment of each group. Blood glucose levels of the test animals were measured at 0 minutes before treatment and 30, 60, 90, and 120 minutes after induction. Data on blood glucose levels each time was converted into data AUC0-120 and the percentage of hypoglycemic effect. Statistical analysis using ANOVA followed by Duncan's test at a 95% confidence level. The results showed that the water fraction of pepolo stem bark had a hypoglycemic effect on male mice of the Balb/C strain loaded with glucose, with a percentage of hypoglycemic power of (40; 36 and 31)%, respectively. Meanwhile, in animals loaded with sucrose, the percentage of hypoglycemic power was (22, 21, and 13)%. The effective dose for the hypoglycemic effect of pepolo bark extract is 200 mg/kg BW.
